Improvement and evaluation of a rat model of postpartum depression induced by hormone withdrawal following hormone-simulated pregnancy

摘要: 目的 改进激素模拟妊娠后激素停撤大鼠产后抑郁模型的建立方法,并对模型进行评价。 方法 25 只 SD雌性大鼠随机分为正常组(10 只)和实验组(15 只) 。 实验组去势手术后筛选合格动物,前 16 d 每日皮下注射雌二醇 2. 5 μg,黄体酮 4 mg,后 7 d 每日注射雌二醇注射液 50 μg。 正常组不做手术处理,23 d 持续注射等量橄榄油。建模后第 1 周通过旷场实验、强迫游泳实验进行大鼠行为学检测。 结果 与正常组相比,实验组旷场实验水平运动距离减短,强迫游泳实验不动时间延长,差异均有统计学意义( P< 0. 05) 。 结论 通过改进建模方法,成功建立稳定的产后抑郁大鼠模型,并探寻到可靠的评价方法。

Abstract: Objective To improve the method of establishing a postpartum depression rat model after hormone withdrawal following hormone-simulated " pregnancy" and evaluate the model. Method 25 SD female rats were randomly divided into normal group ( n = 10) , experimental group ( n = 15) . Qualified animals were selected after ovariectomizing bilaterally in the experimental group, which received daily injections of the hormones estradiol 2. 5 μg and progesterone 4 mg for the first 16 days, estradiol 50 μg for the next 7 days. In the normal group, no treatment was performed, and the same amount of olive oil was injected for 23 days. In the first week after modeling, the behavioral tests were conducted through Open Field Test ( OFT) and Forced Swim Test ( FST) . Result Compared with the normal group, the experimental group exhibits significantly shortened horizontal movement distance in the OFT and increased immobility time in the FST. The difference was statistically significant (P<0. 05) . Conclusion By improving the method of establishing model, a model of postpartum depression rat after hormone withdrawal following hormone-simulated " pregnancy" was successfully established, and a reliable evaluation method was found.
